骨吸收標志物CTXI的檢測
CTXI是I型膠原交聯羧基端肽,Ⅰ型膠原由三條肽鏈組成,其中有兩條α(Ⅰ)鏈,一條 α(Ⅱ)鏈,成熟的I型膠原分子的羧基端具有呈非螺旋的 3 條較短的肽鏈結構,CTXI是 I 型膠原在降解的時候從末端解離下來的肽段。由于膠原蛋白獨特的三股超螺旋結構,性質十分穩定,一般的加工溫度及短時間加熱都不能使其分解,從而造成其消化吸收較困難,不易被人體充分利用。
I 型膠原主要分布于皮膚、骨、角膜、肌腱、腫瘤中,CTXI作為Ⅰ型膠原的降解指示劑,以完整的免疫原性蛋白形式進入血液中,不再進一步分解,故可作為骨吸收標志物,主要用于監測絕經后婦女和骨量減少個體的抗骨吸收治療,評價患骨代謝疾?。ㄈ绺奔谞钆韵俟δ芸哼M、佩吉特病、骨營養不良),在局部骨溶解增加的狀態下,如多發性骨髓瘤、溶骨性骨轉移、類風濕性關節炎及固定術中,可觀察到血清CTXI上升,同時CTXI對于乳腺癌骨轉移早期診斷有一定價值。
由于這個端肽表達量較低(在人血清樣本中濃度為142.6~1638.9pg/ml,在小鼠血清樣本中濃度<545.8pg/ml ),故而研究難度較大,市面產品少。為了滿足研究人員對其的研究需要,Cloud-Clone針對性的開發了CTXI端肽、抗體以及檢測試劑盒等。端肽(SPA665Hu01、CPA665Hu21、CPA665Hu11、CPA665Hu31)、單克隆抗體(MAA665Hu21)和多克隆抗體(PAA665Hu01、LAA665Hu91和LAA665Hu71)的特異性都有實驗數據支撐。檢測試劑盒可定量測定人血清、血漿或其他相關生物液體中CTXI含量(CEA665Hu),Cloud-Clone還開發了小鼠(CEA665Mu)及大鼠(CEA665Ra)CTXI的檢測試劑盒,以滿足不同研究者的需要。
